Semaglutide
Also indexed as Ozempic · Wegovy · Rybelsus · GLP-1 receptor agonist
Semaglutide is a long-acting GLP-1 receptor agonist engineered from the native hormone pathway. The same molecule appears in several approved products, but product, indication, label and route still matter.

O que aparece na literatura
It activates the GLP-1 receptor, a hormone receptor involved in glucose-dependent insulin secretion, appetite signaling and gastric emptying. Those are high-level pharmacology concepts, not instructions for use.
History & development
- 2002–2010s
Novo Nordisk's peptide-engineering program worked through many candidates to extend GLP-1 activity. The company history identifies semaglutide as compound 217 and describes fatty-acid albumin binding as the key half-life strategy.

- 2017
The FDA approved Ozempic (semaglutide) for adults with type 2 diabetes under its labeled indication.
- 2019
Rybelsus became the first oral GLP-1 product for type 2 diabetes in the Novo Nordisk heritage timeline; PIONEER 1 is a published randomized study in that program.

O que a fonte sustenta
Randomized clinical weight-loss research
STEP 1 studied once-weekly semaglutide plus lifestyle intervention in adults with overweight or obesity and reported substantial weight loss at 68 weeks.
Limite: The population, dose, product, follow-up and protocol were specific; this is not a marketplace-material assessment.

Cardiovascular outcomes trial signal
SELECT evaluated semaglutide in adults with overweight or obesity and established cardiovascular disease without diabetes, with a lower incidence of major adverse cardiovascular events.
Limite: An outcomes result in a defined population is not a general prevention promise or a quality claim about an unverified product.

Longer-duration clinical research association
STEP 5 reported two-year weight and safety outcomes under a defined randomized extension protocol.
Limite: Product, adherence and protocol were study-specific; persistence of a study signal is not a product-quality claim.

Randomized treatment-withdrawal research
STEP 4 randomized adults after an initial semaglutide run-in and showed different weight trajectories after continued treatment versus withdrawal.
Limite: Run-in design and treatment context matter; this does not predict an individual response or support unsupervised use.

Randomized head-to-head clinical research
STEP 8 compared once-weekly semaglutide with daily liraglutide and placebo in adults with overweight or obesity under a defined protocol.
Limite: Head-to-head results still depend on the study population, regimen, adherence and approved clinical context.

Systematic review of randomized trials
A systematic review and meta-analysis can help readers locate adverse-event patterns across randomized semaglutide trials in type 2 diabetes.
Limite: Pooled estimates do not remove individual contraindications, monitoring needs or product-quality questions.

Grades describe the cited record for a narrow question. They do not score a marketplace material or make a health decision.
Naming curiosities & useful distinctions
- The name semaglutide covers a molecule; Ozempic, Wegovy and Rybelsus are product names with different labels.
- Novo Nordisk's own account calls semaglutide compound 217 after 216 earlier candidates.
Compound 217
The number is a development notebook detail from Novo Nordisk's account, not a potency score. It is a useful reminder that a final drug name hides a long candidate-selection process.
One molecule, several labels
The approved products are a regulatory literacy case: brand, formulation, indication and label are part of the evidence record.
